Ampicilline is een aminopenicilline welke een bactericide werking uitoefenen via interferentie met de synthese van de bacteriële celwand. Gram+ spectrum gelijkaardig aan dat van Penicillin G. De toevoeging van een aminogroep verhoogt de werking tov penicilline tegen over Gram- bacteriën echter significant.<ref>{{:ISBN 9780813820613}}</ref>Ampicilline is actief tegen alpha- and ß-hemolytische streptococci, inclusief ''Streptococcus equi'', non–penicillinase-producerende staphylococcen, sommige ''Bacillus anthracis'' stammen, de meeste ''Clostridia'' stammen, vele ''E. coli'', ''Salmonella'', en ''Pasteurella multocida''.<ref name=aavpt/>

Aminopenicillines zijn inactief tegenover ''Pseudomonas'', ''Klebsiella", ''Proteus'', ''Bacteroïdes fragilis'', en penicillinase-producerende staphylococcen.<ref name=aavpt>{{#pmid:14651045}}</ref>
